FOXO4-DRI: Senolytic Peptide Research and Cellular Senescence

FOXO4-DRI (Proxofim) is a retro-inverso peptide that has attracted significant research interest since a landmark 2017 paper by Peter de Keizer and colleagues demonstrated its senolytic properties in mouse models.

Cellular Senescence

Senescent cells — cells that have permanently exited the cell cycle — accumulate with age and contribute to tissue dysfunction through the senescence-associated secretory phenotype (SASP). Clearing senescent cells (senolytics) has emerged as a major focus of aging biology research.

FOXO4-p53 Interaction

Senescent cells resist apoptosis partly through an interaction between FOXO4 and p53. FOXO4-DRI is designed to interfere with this interaction, thereby restoring apoptotic sensitivity specifically in senescent cells.

Research Findings

The de Keizer study demonstrated that FOXO4-DRI treatment in naturally aged mice led to improved physical fitness, fur density, and renal function. Independent replication and human studies remain priorities for the field.
